направленное выключение экспрессии определенного гена (нокаут) за счет превращения его в нефункционирующую форму с помощью трансгеноза и гомологичной рекомбинации, которое в отличие от обычного нокаута гена (см. гена нокаут) осуществляется или на определенных стадиях развития организма, или в определенных типах клеток (тканей). Для осуществления О.у.н.г. существует несколько методических подходов. Один из них заключается в следующем. Первоначально получают 2 вида трансгенных мышей:1) мышь, содержащую вместо целевого нормального гена искусственно вставленный тот же ген, который фланкирован с двух сторон специфическим вирусными элементом loxP-сайтом;2) мышь, содержащую вирусный ген cre (кодирует фермент, осуществляющий рекомбинацию по loxP-сайтам), находящийся под контролем определенных ткане- или стадиеспецифических регуляторных элементов. Далее для проведения О.у.н.г. требуется скрещивание этих двух видов трансгенных организмов. У двойных трансгенных мышей происходит вырезание целевого гена под действием рекомбиназы Cre, но только в тех тканях или на тех стадиях развития, когда ген cre экспрессируется.Толковый биотехнологический словарь.
